제목 Target Specific Intracellular Delivery of anti-VEGF siRNA Using Hyaluronic Acid Reducible PEI [HA-(PEI-SS)] Conjugate
초록 Target specific intracellular siRNA delivery systems have been developed using hyaluronic acid (HA) reducible polyethyleneimine (PEI) conjugate. To alleviate the inherent cytotoxicity and low serum stability of high molecular weight (MW) PEI, relatively non-toxic low MW PEI was crosslinked with cystamine bisacrylamide (CBA) to synthesize reducible PEI-SS in the cytosol. Then, PEI-SS was conjugated with HA in the forms of HA-g-(PEI-SS) and HA-b-(PEI-SS) to enhance serum stability and facilitate target specific delivery of siRNA by receptor. The HA-(PEI-SS) conjugates could make a complex with siRNA showing negative surface charge. The resulting HA-(PEI-SS)/siRNA complex demonstrated an excellent gene silencing efficiency. Moreover, the cytotoxicity of HA-(PEI-SS) was negligible likely due to the degradation of PEI-SS. In vivo gene silencing tests using anti-VEGF siRNA will be carried out for the treatment of various angiogenic diseases including cancer and asthma, and liver cancer.
