Target specific intracellular delivery of small interfering RNA (siRNA) has been one of the most important barriers for the development of siRNA therapeutics. In this work, we successfully developed cysteamine modified gold / siRNA / polyethyleneimine (PEI) / hyaluronic acid (HA) complex using layer-by-layer method for target specific intracellular delivery of siRNA by HA receptor mediated endocytosis. Zeta potential and particle size analyses confirmed the formation of Au/siRNA/PEI/HA complex having a particle size of ca. 160 nm and a slightly negative surface charge. With negligible cytotoxicity, the Au/siRNA/PEI/HA complex showed an excellent target specific gene silencing efficiency higher than 70% in B16F1 tumor cells with HA receptors in the presence of 50 vol% serum. The novel Au/siRNA/PEI/HA complex, which can deliver siRNA specifically to the cells with HA receptors, will be investigated further for clinical applications. |
