Localized Surface Plasmon Resonance (LSPR) biosensor has outstanding advantages of label-free detection, real-time measurement, high sensitivity and simple fabrication. It have been reported that when LSPR biosensor is realized with optical devices such as an optical fiber (OF-LSPR), its capacity is improved definitely. In this study, we assessed the surface linkers to maximize plasmonic changes which occur when an antigen binds on antibody immobilized on the Au nanoparticles of OF-LSPR biosensor. Thus, we prepared three kinds of linkers (citrate, Mixed SAM, Calixcrown) on Au nanoparticles (~ 50nm). The attachment of Au nanoparticles on sensor surface was characterized by SEM and UV-Vis. The efficiency of antibody immobilization was evaluated by measuring fluorescence of FITC-IgG bound on the Au nanoparticle. Finally, PSA of different concentrations were applied to the optimized linker systems for the further studies. |
