siRNA Hydrogel Microparticles for Gene Silencing |

We demonstrated a novel strategy to prepare biologically active siRNA-based hydrogel structures through self-assembly process between complementary ss sense and antisense siRNA species using Y-shaped siRNA as crosslinkers. The siRNA-based microgels with substantially increased charge density were drastically collapsed to form compact nano-sized complexes upon interacting with a less charged cationic polymer carrier, and the complexes exhibited superb cellular uptake and gene silencing activities. In the near future, nano-sized siRNA hydrogel particles could also be fabricated by using a microfluidic device that allows controlled hybridization between the two complementary ss sense and antisense siRNA species in a small mixing volume. Such functionally active siRNA-based nanostructures hold great potentials as a new class of nucleic acid platform materials for therapeutic applications. |
