We have studied coating on the surface films which can detect biomolecules bound on the chip surface and obtained more reliable signals through the label-free biosensor such as a surface plasmon resonance (SPR) sensor. Immobilization of biological entities on the sensor surface often faces serious problems of denaturalization or uncontrolled interactions of the biomolecules, for example, proteins. We have tried to fabricate porous polyelectrolyte multilayer (PEM) films that might keep the immobilized biomolecules in hydrated forms and, therefore, achieved the proper functions of biomolecules. These porous PEM films were utilized to create metal nanoparticles in their three dimensionally interconnected networks, which provided reactive sites to incorporate proteins. We will also discuss cellular interactions on porous PEMs films. |
