Multilineage differentiation of human bone marrow derived mesenchymal stem cells controlled by fibroblast-derived matrix in alginate gels |

Mechanical properties of alginate hydrogels can be easily modulated to control the fate of stem cells and they have been popularly used as extracellular matrix in tissue engineering. However, due to lack of intrinsic cell-binding domains in gels, an additional chemistry is required to introduce cell-binding motifs, resulting in the obstacle in clinical applications. We prepared a 3D cell culture system using a mixture in alginate hydrogel and fibroblast-derived matrix (FDM) obtained by decellularization process. Therefore, FDM provides not only excellent microenvironment for the cell adhesion but also differentiation of human mesenchymal stem cells (hMSCs) in alginate hydrogels. This study presents a 3D culture system of alginate/FDM matrix and suggests that FDM in the alginate gel can control differentiation of stem cells into specific lineage during the 3D culture. |
