제목 A water-soluble and selective naphthalimide-based fluorescent sensor for detection of hydrogen sulfide in live zebrafish
초록 Hydrogen sulfide (H2S) is an important biological messenger, has recently been recognized as a gasotransmitter. The concentration of endogenous hydrogen sulfide has been proven to hane close relation to particular disease, but few biologically-compatible methods are available for its detection in aqueous solution. Herein, we presented two bright fluorescent probes L1 and L2 that are selective for H2S over cysteine, glutathione, homocysteine and other reactive sulfur, nitrogen, and oxygen species in aqueous solution(PBS buffer, pH7.4). Based on previous reports of using H2S to reduce RN3 to the parent amine moiety, we sought to exploit this chemistry for H2S sensing. By masking a fluorogenic amine as azido group, mild reduction with H2S regenerates the parent amine and results in fluorescence turn-on. The probes are simple in structure, easy to synthesize, stable, amenable to long-term storage, and highly selective for sulfide. Our selective and sensitive probes facilitate imaging of H2S in living cells and zebrafish embryos.
