CdSe/ZnS quantum dots(QDs), promising contrast agent for bio-imaging, are modified by double-layer encapsulation of alkoxysilanes; the inner layer for cytotoxicity barrier, while the outer layer for attaching bio-molecules for disease targeting. First, alkoxysilanes such as n-octyltriethoxysilane, dodecyltriethoxysilane and hexadecyltriethoxysilane were utilized to encapsulate CdSe/ZnS QDs via hydrophobic interaction, forming interdigitated hydrocarbon layer and cross-linking reaction of silanol groups, providing siloxane network structure which might be desirable for barriering cytotoxicity. Next, the outer layer was formed by reacting 3-aminopropyl trimethoxysilanes with the alkoxysilanes of inner layer, which also provide amino functional moiety for linking of bio-molecules for disease targeting. Finally, the double-layer silane encapsulated QDs were characterized by UV-vis, fluorescence spectroscopy and TEM. |
