제목 Preparation of PEI-DSPE cataionic liposomes as a gene delivery carrier.
초록 Cationic polymers and cationic lipids are one of the most investigated the non-viral carriers for DNA delivery. Polyethylenimine (PEI) is good vehicle for transferring gene into the cell1. The advantages of cationic lipids are weak toxicity, reduced immunogenecity and thus safety in their use2. In this study, we developed an effective nonviral gene transfer system by combining the advantages of both cationic polymers and lipids. To synthesize the cationic liposomes, novel cationic lipid (PEI-DSPE) having 1,2-diacyl-sn-glycero-3-phosphoethanolamine (DSPE) and PEI were synthesized. The obtained PEI-DSPE was characterized by 1H-NMR (Figure 1). Cationic liposomes (PDCL) were prepared by PEI-DSPE with different molar ratio from 0 to 30 and the mixture of DSPC, HSPC, and CHOL at a molar ratio of 100, 50, and 30, respectively, with phosphate buffered saline solution (10mM). The average size and zeta potential of liposomes were 218.3 nm and +14.3 mV, respectively (Table 1). The PDCL formed the complex with DNA in various weight ratio. In conclusion, we confirmed that PDCL have the possibility as DNA carrier.
