Design of Multifunctional Degradable Polymeric Nanosystems for Active Tumor-Targeting Drug Delivery |

The clinical success of many chemotherapeutics and biotherapeutics intimately depends on the development of non-toxic and efficient tumor-targeting nano-carrier systems. In the past years, we have designed and developed various types of stimuli-sensitive degradable polymeric nano-vehicles for tumor-targeting drug and protein delivery, which include (i) bio-responsive, in particular reduction- and pH-responsive, degradable micelles and polymersomes for efficient loading and targeted intracellular release of anti-cancer drugs and proteins; (ii) robust and stimuli-sensitive micelles and polymersomes to simultaneously resolve the in vivo stability and drug release dilemma; and (iii) acid-sensitive prodrugs, prodrug nanoparticles, and prodrug nanogels that are sufficiently stable during circulation but activable inside the target tumor cells in response to a low pH in the endo/lysosomal compartments. |
