Evaluation of in vitro biocompatibility and in vivo bone regeneration ability of surface modified hydroxyapatite granular bone filler with Sr and Si doping |

Surface modification was done to Strontium and Silicate-doped hydroxyapatite granular bone filler by coating with collagen and loading with BMP-2 to enhance in vitro biocompatibility and in vivo bone regenerating ability. Sr and Si doped-HAp powder was used to fabricate granular bone fillers with 1 mm in diameter using the fibrous monolithic process. The surface of the sintered granules was modified by coating with collagen and BMP-2 growth factor. SEM, XPS, porosity and mechanical strength analyses were done to evaluate the effect of the modification on the morphology and structure of the granules. MC3T3-E1 pre-osteoblast cells were used to test for quantify cell proliferation by MTT assay and cell adhesion behavior by confocal laser scanning microscopy. Cell differentiation was monitored using western blotting and RT-PCR. For in vivo tests, the samples were implanted femoral head of male New Zealand rabbits for 30 days then extracted to conduct micro-CT analysis and histological staining using Masson’s Trichrome and Villanueva Osteochrome staining methods. |
