Redifferentiation of dedifferentiated chondrocyteson cell-derived extracellular matrix |

The primary focus of this work is to evaluate naturally obtained cell-derived matrices (CDMs) as a physical cue in eliciting the redifferentiation of dedifferentiated chondrocytes under 2D environment. Different cell types-fibroblasts, preosteoblasts, chondrocytes- were cultured in monolayer for a week and then decellularized using a cocktail solution of detergents and enzymes. Each CDM was comprised of many components and their quantitative difference was significant. When dedifferentiated rat chondrocytes (P4) were cultured on each CDM, the gene expression of cartilage-specific markers indicated that chondrocytes grown on specific CDM carried much better sign of redifferentiation as compared to other groups. In addition, Immunofluorescene of type II collagen also exhibited the same outcome. In conclusion, CDMs may provide a favorable surface microenvironment for chondrogenic redifferentiation. |
