제목 | Development of recombinant Pseudomonas putida for 3-HP production from glycerol - Disruption of 3-HP degradation pathway |
초록 | Biosynthesis 3-HP has been developed using recombinant microorganisms. Using Pseudomonas denitrificans for 3-HP production has several advantages, including: (i) aerobically organism; (ii) naturally producing B12 and (iii) 3-HP inducible systems is well-characterized. However, one of disadvantage is this strain only utilize gluconate as carbon source. In attempt to screen various hosts which has potential to produce 3-HP and also can utilize cheap carbon source (such as glucose), Pseudomonas putida could be a potential strain because of its characteristics. In this study, genes related to 3-HP degradation pathway were disrupted by using marker-less chromosomal gene deletion based on sacB selection system. Strain after removal of 3-HP catabolic genes was examined 3-HP degradation with growing cells and resting cells. As a result, this strain could produce 3-HP from glycerol by using plasmid overexpress genes in 3-HP pathway. |
