Tumor Penetrating Peptide Enhances Tumor Uptake of Co-administered Monoclonal Antibody |

A tumor-penetrating peptide, CRGDKGPDC (iRGD) was reported to enhance vascular permeability and tumor tissue penetration of co-administered drugs in a αvβ3 specific and neuropilin-1 dependent manner. We investigated the effect of intravenously co-administered iRGD on the enhancement of tumor vascular permeability to In-111 labeled B3, IgG1k mAb directed against Ley antigen. We used A431 tumor-bearing nude mice as a model. A431 cell expresses Ley, but not αvβ3 and neuropilin-1on the cell surface. The results of the study indicate that the iRGD coinjection increased the extravasation of In-111 B3 into interstitial space of A431 tumor by 29~31%. |
