In order to minimize the electrostatic aggregation between drug delivery carrier and serum proteins during blood circulation, nanocarriers with selective charge converting characteristics was achieved by conjugating 2,3-dimethylmaleic anhydride (DMMA) to the amino group of the octadecyl grafted poly(2-hydroxyethyl aspartamide) (PHEA-g-C18-NH2). PHEA is a biocompatible and biodegradable poly (amino acid) derivative, and DMMA selectively detaches from it when acidic tumor environment is reached, generating a charge converting system. Not only this would preventserum aggregation, but alsowould enhancethe cellular uptake of the anticancer drug. Characterization of the polymeric micelle was fully investigated, with enhanced cellular uptake and anticancer therapeutic effect was evaluated by FACS, CLSM, and MTT assay. |
