제목 Osmotic pump tablet using solid dispersion to control release of Nifedipine
초록 Nifedipine (NIF) which has an effect of hypertension is poorly water soluble and photodegradable drug. The purpose of work is to improve the solubility of NIF by using solid dispersion with squeeze-osmotic pump tablet system (SQZ). A solvent method was employed to prepare solid dispersion system for NIF. HPMC (PVP K25, PEG 6,000) were selected as the water-soluble polymers. After dissolving NIF and HPMC (PVP K25, PEG 6,000) in a suitable volume of an acetone (methanol)/water mixture, this solvent mixture was completely evaporated in a forced-air oven at a temperature of 50-60℃. In this work, another method was used, drug and polymer carriers (HPMC, PEG6000, PVP) are dissolved in methanol to make solid dispersion,. Freeze drying method has been employed to remove solvent (3 days). SQZ composed of one cylindrical squeeze-push layer and drug which located in center of tablet. Tablet was coated with semi-permeable membrane (cellulose acetate). And two orifices were on both side surfaces. During the release test, a 1-ml of water phase (Na2HPO4,SLS and citric acid monohydrate) was pipetted out and kept in an amber auto-sampler vial (1,4,8,12,16 and 24 h). All samples were analyzed with HPLC system within 1 day. In the result, it shows osmotic pump tablet using solid dispersion can improve NIF solubility and release rate.
