Hyperbaric Oxygen-generating Hydrogels |

Oxygen (O2) plays a critical role as a substrate for metabolism and as a signaling molecules regulating cellular activities. In particular, it is demonstrated that hyperbaric O2 facilitates wound healing and vascular development via enhanced cellular activities related to the wound healing process as well as angiogenesis. Here we report hyperbaric oxygen-generating (HOG) hydrogels composed of thiolated gelatin that can form hydrogel networks in situ via calcium peroxide-mediated oxidative crosslinking reaction with O2 generation. We demonstrate that the HOG hydrogels rapidly generate the hyperbaric O2 levels and retain the hyperbaric levels for up to 12 days. The HOG hydrogel enhances the cellular activities of human dermal fibroblasts and endothelial cells related to the wound healing and angiogenesis. We suggest that our HOG hydrogels have great potential as advanced injectable matrices for the treatment of wound healing, vascular disorders as well as in tissue regenerative medicine. |
