Decellularized Heart ECM Hydrogel by Supercritical Carbon Dioxide for Improving the Effect of Angiogenesis |

Decellularization is a removal of cells from tissues and organs, resulting in remaining extracellular matrix components. To preserve useful ECM components and angiogenic cytokines in decellularized extracellular matrix (dECM), decellularization process needs to selectively remove target molecules such as cell membrane and nucleic acid except various ECM proteins and cytokines. Supercritical CO2 (scCO2) is the most common candidate gas because of its low toxicity, low flammability and environmental acceptability. In this study, rat heart tissues were decellularized with scCO2-EtOH and heart dECM was solubilized into acetic acid and formed heart ECM derived-hydrogel. To estimate angiogenesis, heart dECM(HdECM) hydrogel was injected in rat subcutaneous layers and explanted after 3 days. Finally, we suggest that scCO₂-EtOH decellularized tissue could be promising materials for tissue repair and microenvironment around the cells in tissue engineering. |
