We developed calcium phosphate-based nanominerals stabilized by a conjugate of the mussel-inspired adhesive molecule, 3,4-dihydroxy-L-phenylalanine, and a nontoxic hydrophilic natural polymer, hyaluronic acid, for targeted delivery of a therapeutic gene and protein to a desired tissue via a systemic administration. The stabilized CAP nanominerals were able to maintain their colloidal stability in a physiological salt condition for over a week. The ability of CAP nanominerals containing an siRNA or superoxide dismutase to maintain the integrity of the encapsulated biomacromolecules and the stability in the blood allows this formulation to achieve the improved delivery of the therapeutic molecules to the liver and solid tumor after systemic administration. Considering its biocompatibility, transfection efficacy, and tumor targeting capability, this stabilized nanominerals could be considered a promising candidate carrier for systemic cellular delivery of therapeutic genes and proteins. |
