2,3-Butanediol (2,3-BD) is a promising chemical due to its wide variety of applications in the bulk and specialty chemical industries. In this study, Klebsiella pneumoniae KCTC12133BP, which forms few lipopolysaccharide and mucous-like substances, was engineered to enhance the production of 2,3-BD. We aimed to develop several knockout mutants targeted at reducing the formation of by-products with the help of in silico investigation. Although the cell growth and 2,3-BD production rates of the K. pneumoniae ΔldhA ΔadhE Δpta-ackA strain were lower than those of the wild-type strain, the mutant produced a higher titer of 2,3-BD at a higher yield in batch fermentation. In addition, the mutant could produce 91 g/L of 2,3-BD with a yield of 0.45 g/g, with a productivity of 1.62 g/L/h in fed-batch fermentation.
