Most proteins we are interested in have limited information. The actual phenotype is limited because of the complex biological system. Directed evolution can be an alternative to screening mutants by generating random or semi- diversity. The limitation of directed evolution is the size of library can easily exceed the range can be tested. Most products we are interested in are inconspicuous. Therefore, a tool that can screen inconspicuous products in a high-throughput manner is required. In this study, we developed a synthetic screening tool based on transcription factor that reacts with 3-HP. A substrate binding pocket library of ALDH from Azospirillum brasilense was constructed. By using this, we were able to select a strain with 20% increase in yield and productivity of 3HP production compared to the parental strain through a growth-associated screening method, and confirm that the mutant ALDH catalytic efficiency of this strain is increased 3-fold. |
