제목 Photo-crosslinkable and thermo-responsive chitosan/Pluronic hydrogels for gene delivery
초록 A chitosan/Pluronic hydrogel was prepared by a photo-polymerization method for thermo-responsive and controlled DNA release. In particular, Pluronic has been used as injectable hydrogel showing a reversible sol-gel transition behavior at a certain temperature range. Chitosan was employed for a drug reservoir in injectable systems. Chitosan/Pluronic hydrogel system made of di-acrylated Pluronic F127 and glycidyl methacrylated chitooligosaccharide was prepared by varying chitosan contents and different UV exposure time. Released DNA fraction from hydrogel was complexed with chitosan. The effect of chitosan on released profiles was examined by gel retardation and scanning electron microscopy (SEM). Hydrogels with higher chitosan contents well protected DNA molecules compared to those with lower contents. Therefore, the DNA/chitosan complex was expected to exhibit high transfection efficiency.
