Production of biodegradable plastics, P(3HB-co-3HP) from methane |

Polyhydroxybutyrate (PHB) with its biodegradable and biocompatible characters is an attractive polymer. However, its application is limited due to the stiffness and brittleness. Co-polymerisation of 3-hydroxybutyrate (3HB) with another monomer is an alternative to generate biopolymers with more desirable properties. In this study, the pathway for production of poly(3-hydroxybutyrate-co-3-hydroxypropionate) (P(3HB-co-3HP)) in a type II methanotroph was constructed by combining native PHB pathway and 3HP production pathway with the expression of 3HP-CoA synthetase (3HPCS) and PHA synthase. The genes encoding 3HPCS and the PHA synthesis pathway were screened to discover the efficient enzymes for condensing 3HB-CoA and 3HP-CoA into P(3HB-co-3HP). The best recombinant strain OB3b-MCRM-3S was found to synthesize P(3HB-co-3HP) from methane with 9% P3HP in 0.75 g/L CDW. This is the first study reported on production of biodegradable plastics using methane as the sole carbon substrate. |
