Target specific intracellular delivery of siRNA was successfully carried out using polyethylenimine (PEI) - hyaluronic acid (HA) conjugate. The siRNA/PEI-HA complex was formed by electrostatic interaction between siRNA and PEI part of PEI-HA conjugate. The silencing efficiency of luciferase reporter gene and VEGF gene by siRNA/PEI-HA complex was more significant in B16F1 cells with HA receptors than that by siRNA/PEI complex. Furthermore, intra-tumoral injection of anti-VEGF siRNA /PEI-HA complex resulted in an effective inhibition of tumor growth by the HA receptor mediated endocytosis to tumor cells in C57BL/6 mice. The novel PEI-HA conjugate, which can deliver siRNA specifically to the cells containing HA receptors, will be investigated further for clinical applications. |
