Tyrosine residues in macromolecules can be hydroxylated to form dopa which is found in mussel proteins. And the bimimetic application of the mussel protein by introducing dopa residues in various matrixes is broadly introduced in biomedical field, nanotechnology, tissue engineering etc. The dopa moieties can be selectively introduced by converting from tyrosin residues in natural polymers such as gelatin with tyrosinase. We screened tyrosinase that efficiently bio-catalyze tyrosine residue in macromolecule, and used it for introducing dopa moieties to gelatin. The dopa modified gelatin turned out to have a new function that can detect the pH change in the presence of ferric ions; the dopa-ferric ion complex can be form reversely depending on pH and detected visually by its color changes. Since the gelatin itself possesses limiting properties for being applied in various usages, carbohydrate such as hyaluronic acid and carbohydrate was incorporated after chemically modified and cross-linked to gelatin by tyrosinase and laccase. |
