Photosensitizer combined with charge tunable polymer in cancer environment for targeted cancer therapy |
초록 |
Photodynamic Therapy have been used to therapy of various diseases. When photosensitizer is irradiated, Reactive Oxygen Species is generated. Then, ROSs damage to cell membrane. But it has challenges such as low solubility and non-targeting of tumor. To resolve these challenges, PSs require targeting feature, water-solubility. In this study, we synthesized PSs with polymer (SPP). Used polymer have tumor-targeting feature. SPP be stable in the blood vessel due to the negative charge. It is accrued in a tumor by the enhanced permeability and retention effect. Polymer changed positive charge at tumor because polymer is protonated. SPP interacted negative charge of tumor membrane. We characterized using 1H-NMR, fluorescence spectroscopy, dynamic light scattering. In vitro pH-dependent cellular uptake was measured using confocal laser scanning microscopy, flow cytometry. Therapeutic potential was confirmed using an MTT assay |
