Thermogelling Pluronic F127 hydrogels, approved by the FDA, have been extensively studied for drug delivery and tissue engineering. In this study, Pluronic F127 (F) derivatives, F127-glycolide-4-methacryloxyethyl trimellitic anhydride (FGM) or FM, were modified by grafting cell adhesive RGD peptide for the improvement of chondrocyte adhesion. These polymers were characterized by 1H-NMR, FTIR and elemental analyzer. Cell viability of chondrocyte was assessed in a form of hydrogel. Physical stability of F127 derivatives hydrogel was promoted through the treatment of poly(ethylene glycol) diacrylate (PEGDA), whereas cell viability was low due to the cytotoxicity with higher content of APS/TEMED used as a crosslinking agent. Moreover, the pH adjustment of FGM or FM gel using sodium hydroxide had an influence on cell viability and improved cell proliferation. Therefore, we expect F127-based hydrogels grafted with RGD peptides to provide an appropriate environment for 3D cell culture. |
