제목 A novel biodegradable branched poly(ester amine)s based on glycerol dimethacrylate and low molecular weight polyethylenimine as a gene carrier
초록 Gene therapy is a new approach in the treatment of numerous diseases caused by damage or lack of certain genes. Cationic polymers are very promising tools for gene delivery into different cell types, but need to be improved in certain area for success. Polyethylenimine (PEI) is a non-degradable gene carrier with high transfection efficiency and high toxicity as well. We synthesized low cytotoxic biodegradable poly (ester amine)s, as a novel gene carrier. The copolymers were synthesized using low molecular weight branched PEI (1.2 KDa) with a hydrophilic, biodegradable glycerol dimethacrylate through Michael addition as per our previous reports. The complexation of polymer/DNA was characterized for complex forming ability, particle size and morphology. Cytotoxicity and transfection efficiency of copolymers was determined on cultured HeLa, HepG2, and 293T. In vitro transfection assay and cytotoxicity of degradable poly (ester amine)s was quite comparable to that of PEI.
