Pluronic hydrogels, both F68 (MW: 8,400) and F127 (MW: 12,600) were conditionally approved by the FDA as biomaterials for human applications. As compared to F127, Pluronic F68 has a limitation as an injectable gel because it is unable to show a temperature-responsive sol-gel transition, especially at body temperature. Since many studies have focused on the solutions, we have developed novel temperature-sensitive, biocompatible Pluronic F68 derivatives. A cell-recognition motif, Gly-Arg-Gly-Asp (GRGD) peptide was grafted to F68-L-lactide- 4-methacryloxyethyl trimellitic anhydride (FLM) and F68-caprolactone-M(FCLM), respectively. When these functional polymers were characterized by using FTIR, 1H-NMR, GPC, elemental analysis, and dynamic mechanical analysis, the preliminary data suggest that the GRGD-grafted F68 derivatives are possible in controlling the sol-gel transition. |
