Simultaneous control of cellular gene activation and its PEG-based hydrogel microenvironment via conventional LED light |
초록 |
In tissue engineering, providing an optimal cellular microenvironment has been considered to be a key criterion in designing scaffolds. Although incorporating responsive units has been able to develop cell-friendly niche, direct control of the microenvironment was limited. To address the limitation, light responsive linker has been introduced to hydrogel to directly modulate cellular microenvironment. From this approach, simultaneous temporal and spatial control of ECM degradation is possible, ultimately resulting in more precise direction of cellular activities. Additionally light responsive cells have been developed to manipulate cellular functions with a genetically encoded light-switchable transgene system. Finally, we encapsulated cells with light inducible dimerizing proteins for targeted gene control within a photodegradable PEG-based hydrogel scaffolds. Through blue LED light source, efficient gene control and scaffold remodeling system was enabled for tissue engineering. |
