제목 | Photo-mediated TRAIL gene loaded human mesenchymal stem cell for cancer therapy |
초록 | Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a promising therapeutic protein to selectively induce cancer cell apoptosis. Because of denaturation, delivery of TRAIL protein exhibits low stability and short half-life. To deliver the TRAIL effectively to tumor site, human mesenchymal stem cells (hMSCs) were applied for the direct secretion of TRAIL to target tumors and use tumor homing effect. The TRAIL gene loaded stem cell was prepared by using non-viral vectors, but non-viral vectors induced hard to transfection. Thus, photochemical internalization (PCI) was utilized for enhanced transfection efficiency of secreting TRAIL from hMSCs. We demonstrate that PCI-mediated polyplex loading significantly enhanced TRAIL expression from stem cells and that homing ability magnified cancer targeting. Consequently, we implied that the delivery of PCI-polyplex@hMSCs offers exciting potential treatments in pancreatic cancer gene therapy. |
