Thermosensitive poly(organophosphazene) hydrogel having functional groups was conjugated with cell adhesion peptide GRGDS into polymeric backbone. Synthesized poly(organophosphazene)-GRGDS conjugates showed a range of maximum viscosity value (Vmax) from 297.5 Pa.s to 722.5 Pa.s near the body temperature due to difference of hydrophobic interaction of each conjugates. Cell proliferation and differentiation of GRGDS-conjugated hydrogels were evaluated by injection into a nude mouse model using rabbit mesenchymal stem cells (rMSCs). The three experimental time points for the encapsulated cell study were 1, 2, and 4 weeks, with GRGDS-conjugated hydrogels or without GRGDS-conjugated hydrogel. Analysis for cell differentiation was examined by H&E, histology, and immunohistochemical staining. In this study, we synthesized an injectable thermosensitive hydrogel conjugated with cell adhesion sequence of RGD for application in bone tissue engineering. |
