Over the last decade surgical revascularization procedures by using stent have been increasing dramatically, but many patients suffer from reoccurrence of stenosis, restenosis, after implantation of stents. Therefore, it is important to develop the method for preventing restenosis. Nowadays, there are many drug-eluting stents to prevent this phenomenon by reducing the proliferation of smooth muscle cells, but recent strategies are not satisfactory. Nitric oxide (NO) is a highly reactive free radical molecule known for mammalian biological signaling pathway, which is produced by a NOS (nitric oxide synthase) in vivo. This NO has been shown to reduce platelet adhesion and proliferation of smooth muscle cells, but in general method, fast release of NO is a disadvantage for further application. Therefore, in this experiment, we used thermo-sensitive pluronic F127 for controlled NO release by gelation, and non-cytotoxic branched polyethylenimine for NO conjugation. |
