Synergistic Effect of Dual Growth Factor Loading for Enhanced bone Regeneration |

Natural tissue regeneration is a complex process in which responding cells are regulated by the coordinated action of several environmental cues. For bone tissue engineering, the strategy of providing multiple cues may be particularly important because bone formation and repair are regulated by many factors, including specific ECM properties and several growth factors. Current therapies rely on bolus delivery of single growth factor, thus success of clinical trials has been limited. In this study two growth factors were evaluated, a protein growth factor and a transforming growth factor. Biological activity was measured using a cell-based bioassay and biochemical integrity was determined using SDS-PAGE. Cell proliferation, adhesion and attachment of human adipose stem cells observed by confocal microscopy was improved and differentiating hBMSs showed higher mineral deposition and calcium content. We demonstrate that dual delivery of platelet-derived growth factor (PDGF-BB) and bone morphogenetic protein 2 (BMP-2), each with distinct kinetics, from a single structural composite scaffold may result in rapid formation of a mature vascular network and enhanced bone formation. |
