제목 | Preparation of PHBV/Keratin nanofiber mats by electrospinning |
초록 | [Introduction] Keratins are fibrous proteins constructing hair, wool, nail and so on, but they are water-insoluble because of high disulfide bond content. Poly(3-hydroxybutyrate-co-3-hydroxyvalerate)(PHBV) is well known as biodegradable, biocompatible and non-toxic polymeric materials. Based on above properties, we aimed to obtain PHBV/Keratin nanofiber mats by electrospinning, which have potential usages in tissue engineering scaffolds, wound dressing, drug delivery platforms and so on. [Experimental] 1. Modification of keratin After optimizing conditions for extracting keratin from raw materials, we chemical modified cysteine residues with iodoacetic acid (IAA). The degree of modification was measured by SH% assay using DTNB method. 2. Electrospinning of PHBV/keratin Nanofiber mats were obtained by electrospinning of 6 wt% polymer solution in perfluoroalcohol with different ratio. Characterizations of nanofiber mats were carried out using ATR-FTIR, SEM, AFM and ESCA. 3. Bio-related experiments To examine cell behaviors on nanofiber mats, fibroblasts were used. The other bio-related experiments are ongoing. [Results and discussion] Fig.1 shows SEM image of nanofiber mats obtained from PHBV/Keratin blends. Fig. 1.
