Pancreatic cancer is one of the deadliest cancers leading fifth death rate in Korea. 95% of pancreatic cancer is determined as pancreatic ductal adenocarcinoma (PDAC). Not only is it difficult to be diagnosed at early stages but also most cases show poor prognosis. Nevertheless, the physiology of PDAC cell is still veiled. In recent years, it has been shown that 3D culture platform is a promising tool to recapitulate the behavior of PDAC cells compared with conventional 2D TCPs. In this study, PEG-based hydrogels were fabricated by thiol-ene photo-click reaction to render 3D microenvironment for pancreatic cancer cells with different properties (e.g., stiffness, bioactivity, degradability). The apoptosis of PANC-1 cells was enhanced with epidermal growth factor receptor inhibitor in stiff matrix. The chemoresistance of COLO-357 cells increased in 3D hydrogel matrix. These findings suggest PEG-based hydrogel matrix can give us useful solutions to understand physiology of PDAC cells. |
