Cloning and expression of an Arabidopsis glycosyltransferase in various Escherichia coli strains |

Glycosyltransferase(GT) is the enzyme that catalyzes the transfer of glycosyl group to aglycon. GT can be used for the synthesis of various glycosides such as glycosylated flavonoids. In this study, a gene encoding GT was cloned from Arabidopsis and expressed in various Escherichia coli strains to enhance the expression level. Three types of E. coli strains such as Rosetta, BL21 (DE3) and HMS 174 were used as the host cells and evaluated for their glycosylation activities.A variety of flavonoidswere employed assubstrates. The recombinant whole cells and the purified enzymes could be used as the biocatalyst for glycosylation of flavonoid. |
