제목 | Preparation and characterization of solid dispersion of acyclovir with water soluble polymers |
초록 | Acyclovir (ACV) is anti-viral drug(against HSV-1, HSV-2) which has different dissolution behaviors in water at different temperature.(1.3㎍/㎖ at room temperature) This work checked the dissolution behaviors by rising temperature. The Cmax of ACV in water is 40mg/ml (at 80℃). To make solid dispersion, ACV and polymer carrier are solved in water. Spray dry, freeze dry are used making solid dispersion. Morphology and crystallization were obtained from SEM and XRD. Also, we experiment degeneration of ACV at high temperature. ACV solid dispersion which was to put in hard capsule compared with commercial reference drug by dissoluting in gastric juice and intestinal juice.(sampling time 5, 15, 30, 45, 60, 90, 120, and 180 minutes) The dissolution rate of the three commercial reference drugs has low concentration in gastric juice. Behind the back, ACV solid dispersion that we manufactured have more high dissolution behavior. We expect to obtain high bioavailability and this work can reduce side effect by using H2O not organic solvent. Acknowledgement: This work supported by KOSEF(R01-2001-00431) |
