Cellular Internalization of Hydrophobically-Modified Chitosan Nanoparticles: Endocytosis, Exocytosis, Drug Release into Cytosol |

Here we describe self-assembled nanoparticles as a promising system for intracytoplasmic delivery of drugs. At slightly acidic environments like endosomes, the nanoparticles were dissembled due to breakdown of hydrophilic/hydrophobic balance. The cellular internalization and drug release of the self-assembled nanoparticles were investigated by flow cytometry and confocal microscopy. The cellular internalization of the nanoparticles was initiated by nonspecific interaction between nanoparticles and cell membranes. The internalized nanoparticles by the adsorptive endocytosis seem to be either exocytosed or localized in endosomes. Flow cytometry and confocal microscopy showed that the nanoparticles slowly released drugs into cytosol and cell cycle anaylsis demonstrated that paclitaxel-incorporated nanoparticles were effective in inducing arrest of cell growth. |
