제목 Preparation of Biodegradable Polyethylenimine-co- Poloxamer as a Gene Carrier
초록 The fields of non-viral gene therapy are rapidly gaining increased interest owing to the potential advantages of these non-viral carriers from view points of their safety, production, and versatility. In this study, reaction of the polyethylenimine(PEI) to a poloxamer macromer as a nonionic water-soluble polymer was performed to get a non-toxic and biodegradable gene carrier. The molecular weight and degradation of the PEI-co-Poloxamer (PEI-POL) were determined using GPC. The PEI-POL/DNA complex was confirmed using agarose gel electrophoresis and DNase I experiment. The sizes and zeta potentials of the complex were measured by electrophoretic light scattering. The cytotoxicity of PEI-POL in 293T, A549 and HepG2 cells was evaluated. The PEI-POL/DNA complex was transfected into 293T, A549 and HepG2 cells. Fig. 1 shows representative transfection efficiency of PEI-co-POL/DNA complexes on A549 according to the concentration of poloxamer concentration to PEI. The results indicated that the transfection efficiency of the PEI-co-POL was higher than that of PEI 25 K as the control and the transfection efficiency of it was decreased with an increase of poloxamer content in the copolymer owing to increased hydrophilicity.
