Rapid restoration of denuded endothelium reduces restenosis and thrombosis. In this study, we introduced silicone nanofilament (SiNf) to increase surface area and coating capacity for antibody immobilization. Also, we employed anti-CD146 antibody (Ab) for the capture of circulatin endothelial progenitor cells. SiNf modified surface was evaluated by FE-SEM, AFM, water contact angle, ATR-FTIR, and XPS. The modified surfaces were characterized at each step including SiNf formation, aminosilanization, and antibody immobilization. The amount of anti-CD146 Ab immobilized on SiNf surface significantly increased compared with other surface. Anti-CD146 Ab sample captured more cells than bare and anti-CD34 Ab ones under in vitro perfusion system. When stents on which was covered with anti-CD146 Abs anchored SiNf were implanted into porcine coronary arteries, re-endothelialization significantly improved and highly reduced restenosis compared to bare metal stent. |
