Intracellular delivery and localization of cationic liposomes containing PEI-DSPE for intratumoral injection of chemotherapeutic agent |

PEI-DSPE-incorproated liposomes (PD-liposomes) were prepared for intratumoral injection of chemotherapeutic agent. The PEI was directly conjugated with phospholipid such as DSPE via amide linkage using EDC and NHS. The PD-liposomes size was 130 ± 10 nm. Additionally, the zeta potential of the PD-liposomes increased from -25 mV to 30 mV by incorporation of cationic PEI on the liposomal membrane. Transfection efficiency of the PD-liposomes to tumor cell was higher than that of the control liposomes, and this result may be explained by electrostatic interaction between partially negative tumor cell and positive PD-liposomes. Notably, localization of the PD-liposomes at tumor tissue was observed by fluorescence microscopy and the PD-liposomes showed higher localization than the control liposomes and the free calcein. In conclusion, the PD-liposomes are potent carriers to enhance cell transfection and intratumoral localization of chemotherapeutic agent. |
