Preparation of PEG-coated lipid nanoparticles containing amphotericin B and their in vitro and in vivo characteristics |

Lipid nanoparticles (LNP) are drug delivery carriers due to their solubilizing capability and toxicity reduction of poorly water-soluble drugs. Clinical usage of Amphotericin B (AmB), a polyene antibiotic, is limited due to its nephrotoxicity. In this study AmB was entrapped to LNP composed of phospholipid and cholesterol. PEG-coated LNP containing AmB were prepared by incorporating a DSPE-mPEG into lipid composition of LNP. Mean particle size of AmB-entrapped LNP was about 65±7 nm. Surface charge of the AmB-entrapped LNP was negative showing -54.2±4 mV of zeta potential value. AmB entrapment efficiency to LNP reached up to 85±4%. AmB-entrapped LNP were prepared by freeze-drying method. Mean particle size of the AmB-entrapped LNPs was determined before and after freeze-drying to evaluate the LNP stability. In order to investigate effects of PEG on in vitro toxicity of AmB-loaded LNP, cytotoxicity against human kidney cells and hemolysis of red blood cells were evaluated. |
