Tumor vasculature targeting using thiol-reactive PEG-PLLA micelles |

The functionalization of polymeric micelles with tumor-homing peptides enables them to achieve the enhanced therapeutic efficacy against tumors. Herein, vinylsulfone-terminated PEG-PLLA (VS-PEG-PLLA) was synthesized as a thiol-reactive amphiphilic block copolymer and VS-PEG-PLLA micelles revealed the size around 35 nm with a spherical shape. The reactivity of micelles toward thiol-contained targeting ligands (CREKA and cRGDfC) was evaluated by monitoring thiol concentration with Ellman’s method. For tumor vasculature targeting, single or dual ligands-tethered PEG-PLLA micelles were prepared and their in vivo targetability was investigated by optical imaging. |
