제목 | A biodegradable nanofiber scaffold by electrospinning and its potential as wound dressing |
초록 | The blends of PHBVs and proteins were dissolved in a polar solvent and subsequently electrospunned to produce non-woven nanofiber mat1. Polymer solution was prepared by dissolving PHBV in fluoroethanol, and the solution was then transferred to a syringe needle connected to a high voltage power supply. Upon applying a high voltage, a fluid jet was ejected from the needle. As the jet accelerated towards a grounded collector, the solvent evaporated and a charged polymer fiber was deposited on the collector in the form of a non-woven fabric. Circular nanofiber scaffolds were fitted in 24 well culture dish and subsequently immersed with a DMEM medium containing 10% fetal bovine serum. To seed the cells, 200㎕ of NIH 3T3 cell suspension was added and incubated in a humidified atmosphere of 5% CO2 at 37℃ for a requisite time. After incubation, the supernatant was removed, washed twice with a PBS, and fixed by glutalaldehyde aqueous solution. The sample sheet was then dehydrated, dried in a critical point dried, and finally sputter-coated with platinum. These samples were then observed with a field emition scanning electron microscope. The cell viability and proliferation were investigated using MTT and BrdU assay, respectively. In order to examine the biodegradability of the nanofiber mats, samples of 1.5 × 2.0cm size were incubated in duplicate at 37℃ in 6well plate containing DMEM medium in the presence of Peudomonas stutzeri BM310 depolymerase2 or Type I collagenase.
