To determine the optimal conditions for fabrication of cytochrome c thin films, cytochrome c was mixed with the phosphate buffer solution with a various pH and was mixed with ethanol and deionized distilled water (DDW) [2:1:2 (V/V/V)]. After spreading on subphase, cytochrome c thin film was deposited onto the substrate by the Langmuir-Blodgett (LB) technique. The pressure-area (p-A) isotherms of cytochrome c Langmuir films at those pHs were obtained. So, we observed the typical p-A dynamics of cytochrome c Langmuir monolayers. To investigate the cytochrome c LB layers onto substrate, UV-visible absorption spectrum was measured. The cytochrome c molecules have 410nm absorbance. The UV-absorption spectrum of cytochrome c LB layers formed with various pH showed similar maximum absorbance peak. It represents that the cytochrome c molecules were not denatured significantly in those pH.The optimal conditions of film formation with various dipping pressure was investigated. Also, the morphology of cytochrome c LB film onto the substrate was obtained by atomic force microscopy (AFM) |
