제목 Antioxidant copolyoxalate prodrug nanoparticles for the treatment of inflammatory diseases
초록 We developed fully biodegradable vanillyl alcohol-incorporated copolyoxalate (PVAX) that covalently incorporates vanillyl alcohol and peroxalate linkages in its backbone. PVAX was designed to react with H2O2 and hydrolytically degrade, leading to H2O2 scavenging and release of therapeutic vanillyl alcohol. PVAX was formulated into nanoparticles which had a mean size ~550nm in diameter. The polymer nanoparticles are able to image H2O2 and serve as therapeutic agents with potent antioxidant activity. The in vivo studies using a Hindlimb ischemia/reperfusion model showed that PVAX nanoparticels exerted antioxidant and anti-inflammatory activities by reducing the recruitment of eosinophils. Furthermore, PVAX nanoparticles are capable of imaging H2O2 in vivo with excellent specificity over other ROS. PVAX-based chemiluminesent and antioxidant nanoparticles have great potential as a theranostic agent for H2O2-associated inflammatory diseases.
