Phospholipase A1 (PLA1) is an enzyme that has specificity at the sn-1 position of phospholipid. It has many applications in such fields as food, pharmaceutical, and biofuel industries. In particular, in biomass and biofuel industries, PLA1 could be used for degumming of crude oil from plants, which is an essential process for biodiesel production. PLA1 offers a more environment-friendly and efficient alternative to the conventional acid or alkaline catalysts. However, production of recombinant phospholipases have been problematic due to their toxicity against host cells. In this study, we successfully produced functional PLA1 from Serratia sp. in a cell-free protein synthesis system as a cell viability-independent platform for production of cytotoxic proteins. Cell-free synthesized PLA1 was verified to catalyze degumming of crude sesame oil. Cell-free synthesis reaction was also able to be scaled-up without compromising the productivity of functional PLA1. |
