In Vivo Evaluation of PCL Particle-Loaded PLGA/Pluronic F127 Gel as an Injectable Bulking Agent |

Urinary incontinence causes serious medical and social problems, having a deep influence on a patient's psychological state and causing a series of direct social impacts. The causes of stress urinary incontinence have been understood urethral hypermobility or intrinsic sphincter deficiency. Recently, injectable bulking agents have been used as a first therapy for the treatment of stress urinary incontinence due to its relative non-invasion compared to the surgical repairs such as colposuspension or sling techniques. In our previous study, we developed polycaprolactone (PCL) particle (diameter, 100~200 ㎛)-loaded poly(DL-lactic-co-glycolic acid) (PLGA) gel system as an injectable bulking agent to prevent particle migration and volume decrease in the applied site. The PCL particles sized 100~200 ㎛ seemed effective for the volume retention as well as the migration prevention of the particles. In this study, PCL particle-loaded PLGA/Pluronic F127 mixture gel as well as PCL particle-loaded PLGA without F127 were subcutaneously applied to the back of hairless mouse by injection. Pluronic F127 which is hydrophilic and biocompatible was introduced to improve tissue infiltration into the PLGA gel system. The volume change and tissue infiltration into the PCL particle-loaded PLGA gels (with and without F127) were compared with PLGA and PLGA/F127 gels without PCL particles to investigate their performance as bulking agents. |
