제목 Development of Staining Methods for TEM Observation
초록 Staining mechanism of polymers by the staining agents of OsO4, RuO4 were studied. In order to determine the optimum conditions of staining for TEM observation polymer thin films were stained by changing the staining conditions such as concentration of staining agents, staining periods, temperature of samples. Cross-sections of stained bulk samples were prepared by ultramicrotomy in order to examine the thickness of stained region. It was found that the depth of staining agents diffused into polyisoprene by OsO4 reached to ∼300 nm that is approximately 10 times larger than that of polystyrene by RuO4. Degree of staining measured by quantitative analysis of the metal elements from EDX spectra of the stained polymers showed drastic change of staining ability by the functional groups in polymers.
